CDKN1A
Overview
CDKN1A encodes p21, a cyclin-dependent kinase inhibitor and p53 effector that enforces G1/S cell-cycle arrest. It is recurrently altered in urothelial carcinoma.
Alterations observed in the corpus
- CDKN1A was among the genes recurrently altered in FGFR3-altered urothelial carcinoma in the MSK bladder_msk_2023 cohort PMID:37682528.
- Shared trunk mutation in a representative phylogenetic example of bladder cancer primary-metastasis pairs PMID:36543146
- p21 protein induced by activated NOTCH1 (ICN1), mediating G1 arrest and senescence in OSCC cells PMID:23619168
- Mutated in 14% of muscle-invasive bladder carcinomas (BLCA, n=131); predominantly null/truncating mutations indicating loss of function; encodes p21CIP1 cyclin-dependent kinase inhibitor; one of 9 genes not previously reported as significantly mutated in any cancer before this TCGA study PMID:24476821
- H. pylori upregulates histone demethylase KDM4B and drives H4 acetylation at the CDKN1A (p21) promoter, activating PTGS2 and contributing to gastric carcinogenesis PMID:24816255
- p53–p21–RB1 pathway intact in EWS::FLI1-expressing heMSCs; p21 (CDKN1A) induction limits long-term in vitro culture of transduced cells in Ewing sarcoma cell-of-origin model PMID:25186949
- Cell-cycle/RB axis alteration in HCC; part of CDKN2A/CDKN1A/RB1/CCND1/CCNE1 cluster; CDKN2A loss enriched in alcohol-related HCC and independently associated with poor survival PMID:25822088
- CDKN1A A45fs truncal frameshift mutation shared between plasmacytoid and adjacent urothelial NOS components in a mixed bladder tumor, identified by multi-region sequencing PMID:26901067
- Cell-cycle gene alterations including CDKN1A increased significantly with stage (p = 0.028) and grade (p = 0.009) across 105 nonmuscle invasive bladder cancer (BLCA) tumors; TP53/MDM2 and cell-cycle pathway alterations (RB1, CCND1, CDKN1A, CDKN2A) co-enriched with stage PMID:28583311
