CUL3
Overview
CUL3 (Cullin 3) encodes a scaffold protein of the Cullin-RING E3 ubiquitin ligase complex that mediates ubiquitin-dependent proteasomal degradation of target substrates. In lung squamous cell carcinoma, CUL3 functions as part of the NFE2L2/KEAP1/CUL3 oxidative stress pathway: CUL3 forms an E3 ligase complex with KEAP1 to ubiquitinate and degrade NFE2L2 (NRF2). Loss of CUL3 through deletion or mutation impairs NFE2L2 degradation, resulting in constitutive NRF2 pathway activation analogous to NFE2L2-activating or KEAP1-inactivating mutations.
