DLC1

Overview

DLC1 (deleted in liver cancer 1) encodes a RhoGAP-domain tumor suppressor that is frequently deleted in hepatocellular carcinoma. Its chromosomal locus is a GWAS susceptibility locus with SNPs associated with HBV- or HCV-related HCC risk, in addition to its role as a somatic tumor suppressor.

Alterations observed in the corpus

  • GWAS susceptibility locus/SNP associated with HBV- or HCV-related HCC risk; identified as one of several germline susceptibility loci in a 2014 comprehensive HCC genomics review PMID:24735922

Cancer types (linked)

  • HCC: germline susceptibility SNP associated with viral hepatitis-related HCC; historically characterized as a somatic deletion-based tumor suppressor in HCC PMID:24735922

Co-occurrence and mutual exclusivity

  • No co-occurrence or mutual exclusivity data available in this corpus.

Therapeutic relevance

  • No direct therapeutic relevance reported in this corpus.

Open questions

  • SNP-association studies in HCC suffer from poor control selection, retrospective single-center designs, underpowering, and lack of cross-ancestry validation; the DLC1 association requires independent replication PMID:24735922.
