DNM3
Overview
DNM3 (Dynamin 3) encodes a GTPase involved in membrane tubulation and endocytosis. In sinonasal adenoid cystic carcinoma (AdCC), DNM3 was identified as a novel fusion partner of ESRRG (Estrogen Related Receptor Gamma), producing a chimeric transcript not previously described in AdCC.
Alterations observed in the corpus
- Novel ESRRGex3::DNM3ex14 fusion identified in 1 maxillary sinus metatypical AdCC infiltrating the maxilla and arising in respiratory epithelial adenomatoid hamartoma; the fusion is in-frame and not previously described in AdCC or any other cancer context PMID:24418857
Cancer types (linked)
- Adenoid cystic carcinoma of the sinonasal tract: novel fusion partner in ESRRG::DNM3 chimera (1/88 cases); associated with metatypical histology arising in an unusual hamartomatous background PMID:24418857
Co-occurrence and mutual exclusivity
- The ESRRG::DNM3 fusion is distinct from the canonical MYB::NFIB and MYBL1::NFIB fusions; occurs in a metatypical AdCC without canonical MYB or MYBL1 rearrangement PMID:24418857
Therapeutic relevance
- No targeted therapy data. The functional consequence of the ESRRG::DNM3 fusion is unknown; it is unclear whether it activates ESRRG transcriptional targets or disrupts DNM3 endocytic function PMID:24418857
Open questions
- Single case observation limits functional and clinical characterization; whether ESRRG::DNM3 drives oncogenesis through ESRRG transcriptional activation (analogous to MYB::NFIB) or another mechanism is unresolved PMID:24418857
