FANCC

Overview

FANCC (Fanconi Anemia Complementation Group C) is a component of the Fanconi anemia DNA repair pathway involved in resolving DNA interstrand crosslinks. FANCC mutations are associated with improved immunotherapy survival in metastatic urothelial carcinoma.

Alterations observed in the corpus

  • FANCC mutations associated with improved ICI overall survival in metastatic urothelial carcinoma when combined with ATM/RB1 alterations (p = 0.045) PMID:36333289
  • Passenger amplification detected in cSCC (cutaneous squamous cell carcinoma) by GISTIC copy-number analysis PMID:25589618
  • FANCC is a member of the FA pathway gene set; homozygous deleterious FANCC events classify mCRPC patients into the DNA-repair-defect group associated with significantly longer carboplatin response (log-rank P = 0.02) PMID:26928463
  • Combined ATM/RB/FANCC alteration signature present in 73.3% of pre-chemotherapy urothelial carcinoma tumors but only 37.9% of post-chemotherapy tumors (p=0.05), supporting selective elimination of this signature by cisplatin-based chemotherapy PMID:27749842
  • Less frequent DDR alteration in high-grade NMIBC, contributing to the 30% DDR-altered fraction of high-grade disease PMID:28583311
  • One pLoF LP/PV identified by WES of 25 HBOC-related candidate genes in 372 pediatric cancer patients; burden OR=1.3, p=0.534 (non-significant single-cohort). PMID:29489754

Cancer types (linked)

  • Urothelial carcinoma (UC): FANCC mutations part of DDR gene set associated with ICI benefit PMID:36333289

Co-occurrence and mutual exclusivity

  • FANCC mutations co-occur with ATM and RB1 mutations as a biomarker cluster for ICI survival benefit in metastatic UC PMID:36333289

Therapeutic relevance

  • ATM/FANCC/RB1 mutation cluster associated with improved ICI overall survival in metastatic UC PMID:36333289

Open questions

  • Whether FANCC mutation is independently predictive of ICI benefit or acts only in combination with ATM/RB1 alterations.
