FOS
Overview
FOS encodes the c-Fos transcription factor, a component of the AP-1 complex (FOS/JUN heterodimer) that regulates genes involved in proliferation, differentiation, and apoptosis. In cancer, FOS is implicated as a downstream transcriptional target of various oncogenic signaling pathways.
Alterations observed in the corpus
- Elevated mRNA expression observed in GTF2I-mutant thymomas (types A/AB), consistent with TFII-I (GTF2I-encoded protein)-driven transcriptional regulation downstream of the hotspot p.Leu404His mutation. PMID:24974848
Cancer types (linked)
- THYM (thymoma): FOS is upregulated in indolent GTF2I-mutant thymomas (types A/AB), suggesting a role as a downstream transcriptional effector of the TFII-I oncogenic pathway. PMID:24974848
Co-occurrence and mutual exclusivity
- FOS overexpression co-occurs with GTF2I p.Leu404His mutation in indolent thymomas; functional relationship proposed via TFII-I-driven transcriptional regulation. PMID:24974848
Therapeutic relevance
- No direct therapeutic targeting of FOS reported in this corpus.
Open questions
- Whether FOS upregulation is a direct transcriptional target of mutant TFII-I or a secondary consequence of altered TFII-I activity remains to be mechanistically established.
