PDGFRB
Overview
PDGFRB (Platelet Derived Growth Factor Receptor Beta) encodes a receptor tyrosine kinase activated by PDGF-B and PDGF-D ligands, with established roles in mesenchymal and stromal cell biology. In neuroblastoma, PDGFRB marks the mesenchymal stromal (MSC) compartment and distinguishes it from the undifferentiated tumor cluster.
Alterations observed in the corpus
- Expression marker in neuroblastoma: PDGFRB is significantly higher in the mesenchymal stromal (MSC) nC1 cluster than in the undifferentiated nC3 cluster in a single-nuclei RNA-seq study of 11 neuroblastoma tumors (Smart-Seq2), distinguishing stroma from undifferentiated tumor cells. PMID:34493726
- High PDGFRB expression cited as a rationale for sunitinib therapy in metastatic NEN patients PN2 and PN10 in a WGTA precision-oncology study PMID:24326773
- Multi-kinase TKIs active in NPC (cabozantinib, apatinib) have activity against PDGFR family members alongside VEGFR2 and other targets; PDGFRB implicated in angiogenic and tumor-stromal signaling in NPC PMID:24952746
- SMARCC2-PDGFRB fusion identified in B-cell lymphoblastic leukemia/lymphoma (BLL) — TKI target in a pediatric precision-oncology program PMID:28007021
Cancer types (linked)
- NBL — PDGFRB is specifically elevated in the MSC nC1 stromal cluster of neuroblastoma; its differential expression relative to PDGFRA helps distinguish stromal (nC1) from undifferentiated tumor (nC3) populations. High-risk neuroblastomas have significantly higher proportions of stromal cells (FDR <0.01, chi-square). PMID:34493726
Co-occurrence and mutual exclusivity
- PDGFRB is expressed more highly in MSC nC1 than in nC3; PDGFRA is expressed in both nC3 and MSC nC1. The two receptors thus have partially distinct compartmental distributions within neuroblastoma tumors. PMID:34493726
Therapeutic relevance
- No direct targeted therapy is reported in the corpus for PDGFRB in neuroblastoma.
Open questions
- Whether PDGFRB-expressing mesenchymal stromal cells in neuroblastoma actively support tumor growth or are therapeutically targetable is not addressed. PMID:34493726
