RBM10
Overview
RBM10 (RNA Binding Motif Protein 10) is an RNA-binding protein that regulates alternative splicing. It was identified as a novel significantly mutated gene in lung adenocarcinoma, with recurrent truncating mutations suggesting a tumor suppressor role. Its precise mechanism of oncogenesis remains under investigation.
Alterations observed in the corpus
- Truncating mutations (7% of LUAD cases, recurrent nonsense/frameshift/splice events); newly identified significantly mutated RNA-binding protein gene in lung adenocarcinoma PMID:22980975
- Loss-of-function mutations in 8% of LUAD (TCGA, n=230); strongly enriched in males (P = 0.002); 16/21 RBM10 mutations in the transversion-high subset occurred in males (P = 0.003). PMID:25079552
- Loss-of-function mutations detected in 4% of pancreatic ductal adenocarcinoma cases (109-case WES cohort); associated with prolonged overall survival despite aggressive histology, independent of IPMN origin and GNAS status (P=0.0345, n=4 mutant cases). PMID:25855536
- RBM10 observed as a low-frequency hit (>=2 ATC or >=3 PDTC) in a targeted-sequencing study of advanced thyroid cancers (PDTC and ATC). PMID:26878173
- Newly nominated as a significantly mutated gene in colorectal carcinoma (619-case WES cohort); RNA splicing factor; RBM10 mutations previously linked to lung ADC (TCGA 2014) and pancreatic adenocarcinoma. Classified among RNA-processing CRC drivers. PMID:27149842
- Identified as a significantly mutated gene exclusively in lung ADC vs other TCGA tumor types (q < 0.1) in the 1,144-tumor NSCLC landscape study (660 lung ADC, 484 lung SqCC). PMID:27158780
Cancer types (linked)
- LUAD: mutated in 7% of cases (Broad WES cohort, 183 tumors); predominantly truncating mutations PMID:22980975
