SMARCC1

Overview

SMARCC1 (SWI/SNF Related Matrix Associated Actin Dependent Regulator of Chromatin Subfamily C Member 1), also known as BAF155, is a core structural subunit of the SWI/SNF chromatin-remodelling complex. It is recurrently altered in pancreatic ductal adenocarcinoma as part of the broader SWI/SNF disruption landscape affecting over 42% of cases.

Alterations observed in the corpus

  • Part of SWI/SNF chromatin-remodelling lesions present in >42% of pancreatic ductal adenocarcinoma cases (along with SMARCA2, SMARCA4, SMARCC2, ARID1A, ARID1B, PBRM1); specific individual frequency not reported separately. PMID:25855536
  • SMARCC1 recurrently mutated in adenoid cystic carcinoma (ACC) of the salivary gland as a chromatin-regulator mutation alongside SMARCA2 and KDM6A PMID:26862087

Cancer types (linked)

  • PAAD: SWI/SNF complex member recurrently altered; aggregate pathway disruption in >42% of cases. PMID:25855536

Co-occurrence and mutual exclusivity

Therapeutic relevance

  • No direct therapeutic targeting reported; SWI/SNF-deficient tumours may have vulnerabilities to targeted agents (e.g., EZH2 inhibitors in SMARCA4/SMARCB1-deficient contexts).

Open questions

  • Individual mutation frequency and functional role of SMARCC1 alterations specifically in PDA await detailed characterisation.
