WWOX
Overview
WWOX (WW Domain Containing Oxidoreductase) is located at the fragile chromosomal site FRA16D. Loss of WWOX through fragile-site deletions is a recurrent event in esophageal adenocarcinoma (EAC) and other cancers. WWOX functions as a putative tumor suppressor, and its deletion reflects genomic instability in EAC.
Alterations observed in the corpus
- Fragile-site deletion at the WWOX locus is more frequent in EAC compared to CIN gastric adenocarcinoma in the TCGA integrated esophageal/GEA analysis; co-occurs with FHIT fragile-site deletions and elevated in the EAC molecular cluster PMID:28052061
Cancer types (linked)
- EAC: WWOX fragile-site deletions are enriched in EAC relative to CIN gastric adenocarcinoma; the pattern of FHIT/WWOX fragile-site losses distinguishes EAC from other GEA-CIN entities PMID:28052061
Co-occurrence and mutual exclusivity
- Co-occurs with FHIT fragile-site deletions; both FHIT and WWOX deletions are higher in EAC than CIN gastric — a feature noted as potentially distinguishing a clinically meaningful subgroup PMID:28052061
Therapeutic relevance
- No targeted therapy relevant to WWOX loss is reported in the corpus; its significance is primarily as a biomarker of EAC-type genomic instability.
Open questions
- Whether FHIT/WWOX fragile-site loss in EAC defines a clinically meaningful subgroup of CIN GEA requires additional samples to confirm PMID:28052061
