YTHDF3
Overview
YTHDF3 (YTH N6-Methyladenosine RNA Binding Protein F3) is an m6A RNA methylation reader that regulates RNA metabolism. In salivary gland adenoid cystic carcinoma (ACC), YTHDF3 has been identified as an intra-chromosomal fusion partner with MYBL1, with the fusion deleting the C-terminal negative regulatory domain of MYBL1 and driving its overexpression.
Alterations observed in the corpus
- Intra-chromosomal fusion partner with MYBL1 in 2/102 ACC tumors; the MYBL1-YTHDF3 fusion deletes the C-terminal negative regulatory domain of MYBL1 and drives MYBL1 overexpression PMID:26631609
Cancer types (linked)
- ACYC: MYBL1-YTHDF3 intra-chromosomal fusion in 2/102 salivary adenoid cystic carcinomas, part of the spectrum of MYBL1 rearrangements in t(6;9)-negative ACC PMID:26631609
Co-occurrence and mutual exclusivity
- MYBL1-YTHDF3 fusions are part of the MYBL1-rearranged ACC subclass (~14%), mutually exclusive with MYB-NFIB fusions (~53%) and the fusion-negative subgroup (~33%) PMID:26631609
Therapeutic relevance
Open questions
- Whether the YTHDF3 portion of the MYBL1-YTHDF3 fusion contributes oncogenic activity beyond simple MYBL1 derepression is unknown.
