Comet Assay (Alkaline)
Overview
Single-cell gel electrophoresis assay (alkaline variant) that measures DNA strand breaks and alkali-labile sites as a proxy for DNA damage; cells embedded in agarose migrate under an electric field, with damaged DNA forming a characteristic “comet tail.”
Used by
- Alkaline comet assay applied to EWS::FLI1-transduced heMSCs and controls; demonstrated significantly increased basal DNA damage in EF-heMSCs despite elevated BRCA1 levels, supporting a model of defective DNA damage response downstream of EWS::FLI1 expression. PMID:25186949
Notes
- Used alongside phospho-ATM and phospho-ATR western blots in the Ewing sarcoma study to characterize the DDR defect induced by EWS::FLI1.
