AXIN1

Overview

AXIN1 is a scaffold protein in the WNT signaling pathway that facilitates beta-catenin (CTNNB1) phosphorylation and degradation as part of the destruction complex. AXIN1 mutations lead to WNT pathway activation and are observed in gallbladder carcinoma.

  • Loss-of-function mutation in 7% (range 5–10%) of HCCs (WES, n=1,289); WNT–β-catenin pathway component; trunk driver classified as non-actionable PMID:24798001
  • Significantly mutated and recurrently homozygously deleted in HCC; defines its own mutation-association cluster distinct from CTNNB1 PMID:25822088
  • Recurrently mutated WNT-pathway component in PDA; co-mutated with AXIN2 and APC, nominating tankyrase inhibitors (XAV939) as therapeutic candidates PMID:25855536
  • Two ungermlined-paired AXIN1 variants found in the PDTC/ATC cohort (n=117); the study failed to replicate prior reports of frequent WNT-pathway alterations in ATC — these AXIN1 variants did not meet criteria for confirmed somatic WNT-pathway mutations PMID:26878173
