HLA-B

Overview

HLA-B encodes an MHC class I heavy chain that presents peptides to CD8 T cells; loss or mutation of HLA-B contributes to tumor immune escape.

Alterations observed in the corpus

  • HLA-B mutations were observed as part of the subset of private mutations unique to NSCLC brain metastases in the MSK BM cohort PMID:37591896.
  • MHC class I gene highly expressed in HGSOC Cancer.cell.3 cluster; subject to LOH-mediated loss in 6p as immune escape mechanism PMID:36517593
  • MHC class I alteration enriched in MSI gastric adenocarcinoma (STAD) tumours (TCGA); consistent with antigen-presentation escape mechanism; co-altered with B2M PMID:25079317
  • Somatic HLA class I (HLA-A/B/C) mutations identified in 11% of colorectal cancer cases; mutations enriched at TCR-binding and peptide-contact residues; consistent with immune-escape selection in TIL-rich tumors PMID:26997480
  • Implicated in MHC-I antigen presentation loss alongside B2M deletion in TCGA pan-lung cancer cohort (lung ADC and SqCC) PMID:27158780
