IGF2
Overview
IGF2 (Insulin-like Growth Factor 2) is an imprinted growth factor with roles in fetal development and cancer. In rhabdomyosarcoma (RMS), IGF2 is a recurrently altered gene associated with the fusion-positive (FP-RMS) subtype.
Alterations observed in the corpus
- IGF2 is among the recurrent alterations identified in fusion-positive rhabdomyosarcoma (FP-RMS) in a study of 35 tumor pairs using sequential genomic analysis with a combined targeted DNA panel and shallow WGS PMID:37730754.
- Somatic mutations detected in TCGA colorectal adenocarcinoma cohort (276 tumors) PMID:22810696
- Focal amplification and overexpression in 12% (8/68) of colorectal cancers (Genentech WES) PMID:22895193
- IGF2 is a paternally imprinted gene within the 11p15.5 LOH region (50% of RMS tumors); one 3’UTR somatic indel observed; part of the 11p15.5 locus commonly lost in rhabdomyosarcoma PMID:24436047
- ~10% of HCC overexpress IGF2; part of the IGF axis (with IGF2R allelic loss ~25% and IGF1R activation ~21%) targeted therapeutically in HCC PMID:24735922
- IGF2 is epigenetically upregulated and marks the S2 progenitor-cell proliferation subclass in HCC (co-expressed with EPCAM and AFP); proposed as an actionable target in the ~50% of HCCs in the proliferation class. PMID:24798001
- PRC2-repressed imprinted gene aberrantly upregulated in PRC2-loss MPNSTs; promoter H3K27me3 restored after SUZ12 reintroduction in ST88-14 cells with reciprocal reduction of transcript levels PMID:25240281
Cancer types (linked)
Co-occurrence and mutual exclusivity
- Co-occurs with MED12 and NCOR2 alterations as recurrent events in FP-RMS PMID:37730754.
Therapeutic relevance
- No specific IGF2-directed therapy evaluated in this corpus PMID:37730754.
Open questions
- The functional role of IGF2 alterations in FP-RMS progression relative to the dominant PAX3::FOXO1 fusion driver requires further study PMID:37730754.
