NCOR2
Overview
NCOR2 (Nuclear Receptor Corepressor 2, also known as SMRT) encodes a transcriptional corepressor that interacts with nuclear receptors and represses transcription. Recurrent alterations in NCOR2 have been identified in fusion-positive rhabdomyosarcoma (FP-RMS).
Alterations observed in the corpus
- Recurrent alterations in fusion-positive rhabdomyosarcoma (FP-RMS) identified in a sequential multi-platform genomic study of 35 tumor pairs from 27 patients PMID:37730754.
- Recurrently deleted in prostate cancer identified in integrative genomic profiling of 218 tumors PMID:20579941
- Nonsense mutation identified in SHH-subgroup medulloblastoma in WES of 92 tumors (Broad cohort); component of the nuclear co-repressor complex PMID:22820256
- In mCRPC, NCOR2 was identified as an AR pathway regulator recurrently altered in the 150-patient SU2C–PCF prospective cohort. PMID:26000489
- AR co-activator alteration enriched in CRPC-Adeno (21 cases) vs. CRPC-NE PMID:26855148
- NCOR2 is one of only two genes (alongside CSMD1) from 51 metastasis-private non-synonymous variants in patient 99-091 that are altered at >5% frequency in TCGA PRAD or SU2C mCRPC datasets, suggesting most private mutations at metastatic sites are passenger events PMID:26928463
Cancer types (linked)
- Fusion-positive rhabdomyosarcoma (FP-RMS) — recurrent secondary alterations alongside IGF2 and MED12 PMID:37730754.
Co-occurrence and mutual exclusivity
- Co-occurs with IGF2 and MED12 recurrent alterations in FP-RMS PMID:37730754.
Therapeutic relevance
- No direct therapeutic link reported in the corpus.
Open questions
- The functional significance of NCOR2 alterations in FP-RMS progression and their relationship to the PAX::FOXO1 fusion-driven transcriptional program is not characterized.
