P2RY8

Overview

P2RY8 encodes a G protein-coupled purinergic receptor expressed on lymphocytes. Somatic mutations in P2RY8 were identified as recurrent events in diffuse large B-cell lymphoma, suggesting a role in modulating lymphocyte signaling or homing in the tumor microenvironment.

Alterations observed in the corpus

  • P2RY8 harbored 8 somatic mutations in 6 of 55 DLBCL patients by whole-exome sequencing; allelic fractions >0.5 in 3 patients suggested loss of heterozygosity or amplification of the mutant allele PMID:22343534

Cancer types (linked)

  • DLBCL: recurrent somatic mutations, detected in 6/55 (11%) patients in the Broad WES cohort PMID:22343534

Co-occurrence and mutual exclusivity

Therapeutic relevance

  • No drugs directly targeting P2RY8 were reported in the corpus.

Open questions

  • The functional consequence of P2RY8 mutations in DLBCL (gain vs. loss of function, downstream signaling impact) is not established PMID:22343534
