PIK3C2A

Overview

PIK3C2A encodes phosphatidylinositol-4-phosphate 3-kinase catalytic subunit type 2 alpha, a class II PI3K isoform. Unlike class I PI3Ks, class II isoforms preferentially generate PI(3)P and PI(3,4)P2 and are implicated in membrane trafficking, autophagy, and cell migration. PIK3C2A alterations contribute to PI3K pathway activation in a subset of biliary tract cancers.

Alterations observed in the corpus

  • PI3K pathway alteration contributing to a total of 22% PI3K pathway alteration rate in intrahepatic cholangiocarcinoma (IHCH) discovery-screen tumors; identified alongside PIK3CA, PTEN, and PIK3C2G PMID:24185509

Cancer types (linked)

  • CHOL — component of the 22% aggregate PI3K pathway alteration rate in IHCH discovery-screen tumors; exact individual frequency not reported separately PMID:24185509

Co-occurrence and mutual exclusivity

Therapeutic relevance

  • PI3K pathway alterations in IHCH collectively suggest potential sensitivity to PI3K/AKT/mTOR inhibitors, though no specific drug was tested in this study PMID:24185509

Open questions

  • Individual mutation frequencies for PIK3C2A vs. other PI3K pathway members in IHCH were not reported separately; the 22% figure is aggregate.
