PIK3C2G

Overview

PIK3C2G encodes phosphatidylinositol-4-phosphate 3-kinase catalytic subunit type 2 gamma, a class II PI3K isoform. Class II PI3Ks preferentially produce PI(3)P and PI(3,4)P2 and regulate vesicle trafficking, autophagy, and cell survival. PIK3C2G alterations have been identified in biliary tract cancers as part of a broader PI3K pathway activation signature.

Alterations observed in the corpus

  • PI3K pathway alteration contributing to a total of 22% PI3K pathway alteration rate in intrahepatic cholangiocarcinoma (IHCH) discovery-screen tumors; identified alongside PIK3CA, PTEN, and PIK3C2A PMID:24185509
  • Private PI3K-pathway event of unknown significance in paired primary/metastasis CRC cohort; detected in spatially separate tumor regions indicating subclonality PMID:25164765
  • PIK3C2G mutated as part of the PI3K/AKT/mTOR pathway, which is disrupted in 39% ATC vs 11% PDTC (P=1×10⁻³) in a targeted-sequencing study of advanced thyroid cancers. PMID:26878173
  • PIK3C2G S48R mutation observed as a truncal alteration shared between plasmacytoid-variant and adjacent urothelial NOS components in a mixed bladder tumor. PMID:26901067

Cancer types (linked)

  • CHOL — component of the 22% aggregate PI3K pathway alteration rate in IHCH discovery-screen tumors; exact individual frequency not reported separately PMID:24185509

Co-occurrence and mutual exclusivity

Therapeutic relevance

  • PI3K pathway alterations in IHCH collectively suggest potential sensitivity to PI3K/AKT/mTOR inhibitors, though no specific drug was tested in this study PMID:24185509

Open questions

  • Individual mutation frequencies for PIK3C2G vs. other PI3K pathway members in IHCH were not reported separately; the 22% figure is aggregate.
