PTPRD
Overview
PTPRD (protein tyrosine phosphatase receptor type D) encodes a receptor-type phosphatase that functions as a tumor suppressor. PTPRD inactivation through mutations and deletions has been observed in multiple cancer types, including radiation-associated sarcomas.
Alterations observed in the corpus
- Mutations and deletions in 25% of RT-MPNST vs 5% of sporadic MPNST, indicating significant enrichment in radiation-associated malignant peripheral nerve sheath tumors PMID:37350195.
- Frequently mutated and previously shown deleted in LUAD; co-mutates with PIK3C3 in the TSP cohort (n=188). PMID:18948947
- 27 mutations in 17 tumors across protein tyrosine phosphatase receptor domains in the 147-tumor Yale melanoma WES cohort PMID:22842228
- Highlighted in focal-deletion and TieDIE pathway analyses in ccRCC (TCGA 450-case cohort) as a downstream pathway and SCNA feature PMID:23792563
- Recurrent homozygous deletion in multiple myeloma; proposed to feed into IL6/STAT3 signaling as a tumor suppressor through STAT3 dephosphorylation PMID:24434212
- Novel 5’-NFIB fusion partner in adenoid cystic carcinoma; identified in t(6;9)-positive/MYB-NFIB-negative tumors PMID:26631609
- PTPRD tumor-suppressor alterations flagged as actionable in a targeted sequencing study of germ cell tumors, particularly among cisplatin-resistant patients PMID:27646943.
- PTPRD less frequently mutated in the MSK metastatic gastroesophageal cohort vs TCGA non-MSI-H (q<=0.10), suggesting selective pressure against PTPRD mutations in metastatic disease PMID:29122777
Cancer types (linked)
- MPNST – PTPRD mutations/deletions enriched in RT-MPNST (25%) compared to sporadic MPNST (5%) PMID:37350195.
Co-occurrence and mutual exclusivity
- Not specifically reported in the corpus for PTPRD partners PMID:37350195.
Therapeutic relevance
- No PTPRD-directed therapy is reported in this corpus PMID:37350195.
Open questions
- The functional consequences of PTPRD loss in MPNST pathogenesis and whether it cooperates with other enriched RT-MPNST alterations (e.g., NF1, CDKN2A) remain to be determined PMID:37350195.
