RYBP
Overview
RYBP (RING1 and YY1 Binding Protein) encodes a component of the Polycomb repressive complex 1 (PRC1), involved in epigenetic transcriptional repression. Located at 3p14, RYBP is a candidate tumor suppressor in prostate cancer where deletion of this locus is recurrently observed.
Alterations observed in the corpus
- Deletion at 3p14 identified as a candidate tumor suppressor in integrative genomic profiling of 218 prostate cancer tumors PMID:20579941
- Co-deleted with FOXP1 and SHQ1 in a focal 3p13 deletion identified by aCGH and low-pass WGS of FFPE prostatectomy needle biopsies (prad_mskcc_2014 proof-of-principle). PMID:25024180
- Part of 3p21 focal heterozygous deletion (co-deleted with SHQ1 and FOXP1) in MSK-PCa1 and MSK-PCa7 prostate cancer cell lines PMID:25201530
- Part of the complex 3p13 deletion locus involving FOXP1/RYBP/SHQ1 in primary prostate cancer PMID:26544944
Cancer types (linked)
- Prostate adenocarcinoma (PRAD): recurrent deletion at 3p14, co-deleted with FOXP1 and SHQ1 PMID:20579941
Co-occurrence and mutual exclusivity
- Co-deleted with FOXP1 and SHQ1 at the 3p14 locus in prostate cancer PMID:20579941
Therapeutic relevance
- No direct therapeutic targeting currently described; candidate tumor suppressor status warrants further investigation.
Open questions
- Whether RYBP deletion is a driver or passenger event at the 3p14 locus has not been functionally validated.
