SOCS1
Overview
SOCS1 is a negative regulator of JAK/STAT signaling and the most commonly mutated driver in classic Hodgkin lymphoma in the cbio-kb corpus.
Alterations observed in the corpus
- SOCS1 was mutated in 62% of classic Hodgkin lymphoma cases and is the most common cHL driver PMID:36723991.
- SOCS1 showed an elevated noncoding:coding mutation ratio consistent with AID off-target activity PMID:36723991.
- WES of 55 DLBCL tumors identified recurrent SOCS1 mutations, consistent with its role as a negative regulator of JAK-STAT signaling frequently disrupted in B-cell lymphomas PMID:22343534
- Preferentially mutated in GCB DLBCL PMID:28985567
Cancer types (linked)
- CHL — most common driver (62%) PMID:36723991.
Co-occurrence and mutual exclusivity
- Part of the JAK/STAT driver set in cHL alongside STAT6 PMID:36723991.
Therapeutic relevance
- No direct targeted therapy reported in the corpus.
Open questions
- Mechanistic role of AID off-target activity in generating the noncoding SOCS1 mutation footprint PMID:36723991.
