TRAF7
Overview
TRAF7 is a RING domain E3 ubiquitin ligase that is a recurrent driver in the NF2-wildtype (MG2) molecular group of meningiomas.
Alterations observed in the corpus
- TRAF7 point mutations were identified in 25% of MG2 meningiomas (NF2-wildtype group); exclusive to MG2 and mutually exclusive with NF2 mutations PMID:34433969.
Cancer types (linked)
- MNG — point mutations in 25% of the NF2-wildtype MG2 molecular group; part of the TRAF7/AKT1/KLF4/SMO driver set PMID:34433969.
Co-occurrence and mutual exclusivity
- Mutually exclusive with NF2 mutations; co-occurs with AKT1, KLF4, and SMO mutations in MG2 meningiomas PMID:34433969.
Therapeutic relevance
- MG2 tumors harboring TRAF7 mutations may be amenable to targeted therapies directed at downstream effectors; TRAF7 drives NF-kB and JNK pathway activation PMID:34433969.
Open questions
- None flagged in the corpus beyond the need for independent validation of the MG2 molecular group classification PMID:34433969.
