Overview

3-D protein-structure mutation clustering method used to nominate driver genes whose somatic mutations cluster in protein space rather than along the linear sequence.

Used by

  • PMID:35927489 — used in the CLL-map driver-discovery pipeline alongside recurrent sSNV/indel testing; nominated six additional CLL drivers (including MAP2K2, DIS3, DICER1) on top of the 82 from recurrent SNV/indels PMID:35927489.

Notes

  • Specific CLUMPS-nominated driver examples in CLL: MAP2K2 (3 mutations in the kinase domain, activating ERK signaling analogous to MAP2K1), DIS3 (2/4 sites at cancer hotspots D479/D488 in the catalytic domain), DICER1, and INO80 (mutations clustered in the DNA-binding domain) PMID:35927489.
