BIRC2
Overview
BIRC2 (baculoviral IAP repeat-containing 2, also known as cIAP1) is an inhibitor of apoptosis protein (IAP) family member that blocks caspase activation. In OSCC, BIRC2 resides within or near the 11q13 CCND1 amplicon, a region frequently focally amplified in oral squamous cell carcinoma.
Alterations observed in the corpus
- Located within or near the 11q13 CCND1 amplicon in OSCC; copy-number gain with strong CN-expression correlation; possibly contributes to oncogenesis alongside CCND1 in the 22% of OSCC tumors with 11q13 amplification. PMID:23619168
- Within a recurrent significant homozygous-deletion peak in multiple myeloma (MM); part of NF-kB pathway regulatory locus co-deleted with BIRC3; NF-kB pathway alterations retained significance at p<0.05 in the expanded 203-patient MM cohort PMID:24434212
- Inhibitor-of-apoptosis protein cIAP1; targeted by tolinapant + radiotherapy in NPC PMID:24952746
- 11q22 co-amplification with YAP1 in HNSCC (279-tumor cohort); selection inferred from BIRC2-FADD-caspase cascade inhibiting cell death; majority of 11q13-amplified tumors had large telomeric 11q22 deletions including ATM, CASP1, CASP4, CASP5, CASP12 PMID:25631445

Therapeutic relevance
- BIRC2/cIAP1 is a target of SMAC mimetics and IAP inhibitors; amplification in OSCC may indicate context for anti-apoptotic resistance. PMID:23619168
Open questions
- The functional contribution of BIRC2 copy gain to OSCC pathogenesis versus passenger co-amplification with CCND1 at 11q13 is not resolved. PMID:23619168
