Overview

CIC (capicua) is a transcriptional repressor downstream of RTK/RAS/MAPK signaling, recurrently mutated in 1p19q codeleted oligodendrogliomas.

Alterations observed in the corpus

  • CIC mutations observed in 42% of 1p19q codeleted oligodendrogliomas in the MSK-IMPACT subcohort (n=73) of an IDH-mutant low-grade glioma active-surveillance study PMID:37910594.
  • Subclonal loss-of-function CIC mutation (~30% VAF) detected in oligodendroglioma tumor MGH53 by scRNA-seq; mutant cells show increased ETV1 and ETV5 expression consistent with derepression of CIC target genes; mutation found across all three developmental compartments (stem/progenitor, astro-like, oligo-like) PMID:27806376.
