OLIG1
Overview
OLIG1 (oligodendrocyte transcription factor 1) is a bHLH transcription factor required for oligodendrocyte specification and myelination. In oligodendroglioma, OLIG1 marks the differentiated oligodendrocyte-like tumor cell subpopulation, one of two glial lineage programs identified by single-cell RNA-seq.
Alterations observed in the corpus
- OLIG1 is highly expressed in differentiated oligodendrocyte-like tumor cells in grade II oligodendroglioma (IDH-mutant, 1p/19q codeleted), alongside OLIG2 and OMG; these cells coexist with astrocyte-like (GFAP, APOE, SOX9) and stem/progenitor (SOX2/SOX4/SOX11-high) subpopulations within the same tumors PMID:27806376.
Cancer types (linked)
- ODG — OLIG1 marks the differentiated oligodendrocyte-like compartment; used to establish the developmental hierarchy in oligodendroglioma alongside scRNA-seq data PMID:27806376.
Co-occurrence and mutual exclusivity
- Co-expressed with OLIG2 and OMG in oligodendrocyte-like cells; mutually exclusive with astrocyte-like (GFAP, APOE) program at single-cell resolution PMID:27806376.
Therapeutic relevance
- OLIG1-high differentiated cells are non-proliferating (MKI67-low); therapeutic targeting would need to address the MKI67+ stem/progenitor subpopulation rather than the bulk differentiated population PMID:27806376.
Open questions
- Whether OLIG1 expression is causally maintained by the IDH mutation or 1p/19q codeletion in these tumors is not addressed PMID:27806376.
