SOX9
Overview
SOX9 (SRY-Box Transcription Factor 9) is a master transcription factor in chondrogenesis, sex determination, and neural crest development. In glioma, SOX9 marks the astrocytic differentiation lineage and is a key component of the astro-like expression program in oligodendroglioma, defining one of the two major differentiated compartments.
Alterations observed in the corpus
- SOX9 is a defining marker of the astrocytic (astro-like) program in oligodendroglioma (one of 128 astro-high signature genes), co-expressed with APOE, ALDOC, and GFAP, across 4,347 single cells from 6 untreated grade II IDH-mutant 1p/19q-codeleted oligodendrogliomas (Smart-Seq2). PMID:27806376
- Identified as significantly mutated in TCGA colorectal cancer cohort (276 tumors); SOX9 mutations observed among Wnt pathway-related alterations PMID:22810696
- SOX9 alteration drives elevated WNT-pathway mutation frequency in duodenal adenocarcinoma (DUOAC) alongside APC in the pan-periampullary WGS study PMID:26804919
Cancer types (linked)
- ODG — SOX9 marks the differentiated astro-like tumor cells in oligodendroglioma; these cells are predominantly non-cycling and represent one of two glial differentiation endpoints alongside the oligo-like lineage. PMID:27806376
Co-occurrence and mutual exclusivity
Therapeutic relevance
- SOX9+ astro-like cells are largely non-cycling and are unlikely to drive tumor regrowth; therapies targeting the differentiated compartment may be of limited value. PMID:27806376
Open questions
- Whether SOX9 plays a functional role in astro-like differentiation of oligodendroglioma cells or simply marks that state is not addressed. PMID:27806376
