ROBO2
Overview
ROBO2 encodes roundabout guidance receptor 2, a SLIT ligand receptor involved in axon repulsion and cell migration. In pancreatic ductal adenocarcinoma, ROBO2 is mutated in approximately 5% of tumors and low ROBO2 expression associates with poor prognosis, implicating it as a candidate tumor suppressor within the SLIT-ROBO pathway.
Alterations observed in the corpus
- Mutations in approximately 5% of pancreatic cancer tumors in WES of 142 ICGC tumors; low ROBO2 expression associates with poor prognosis in PDAC PMID:23103869
Cancer types (linked)
- Pancreatic ductal adenocarcinoma (PAAD): mutated in ~5% of tumors; low expression is a poor prognostic biomarker PMID:23103869
Co-occurrence and mutual exclusivity
- Co-altered with SLIT2, ROBO1, and ROBO3 as part of the SLIT-ROBO axon guidance pathway in pancreatic cancer PMID:23103869
Therapeutic relevance
- ROBO2 expression level may serve as a prognostic biomarker in PDAC; low ROBO2 expression may identify patients with poor prognosis PMID:23103869
Open questions
- Functional validation of ROBO2 mutations as true drivers in PDAC is incomplete; the prognostic association of low expression requires prospective validation PMID:23103869
