ROBO3

Overview

ROBO3 encodes roundabout guidance receptor 3, a member of the ROBO receptor family. Unlike ROBO1 and ROBO2, ROBO3 acts as an inhibitor of ROBO2 signaling. In pancreatic ductal adenocarcinoma, ROBO3 is overexpressed in some tumors and high ROBO3 expression associates with poor survival, suggesting that ROBO3 overexpression may suppress ROBO2-mediated tumor suppression.

Alterations observed in the corpus

  • Overexpressed in a subset of pancreatic cancer tumors in WES/expression analysis of 142 ICGC tumors; high ROBO3 expression associates with poor survival PMID:23103869

Cancer types (linked)

  • Pancreatic ductal adenocarcinoma (PAAD): overexpressed in a subset; high expression is a poor prognostic biomarker, potentially by inhibiting ROBO2 tumor suppressor activity PMID:23103869

Co-occurrence and mutual exclusivity

  • Functionally linked with ROBO2 (inhibitory relationship); co-altered with SLIT2, ROBO1, and ROBO2 as part of the SLIT-ROBO axon guidance pathway PMID:23103869

Therapeutic relevance

  • ROBO3 expression level may serve as a prognostic biomarker in PDAC; high ROBO3 / low ROBO2 expression pattern identifies poor prognosis patients PMID:23103869

Open questions

  • Functional mechanism by which ROBO3 overexpression promotes poor prognosis in PDAC requires experimental validation PMID:23103869
