MSK-IMPACT 505 (IMPACT505)

Overview

Targeted (505 cancer genes) sequencing of various tumor types via MSK-IMPACT on Illumina HiSeq sequencers — canonical cBioPortal gene panel IMPACT505. The largest/most-recent generation of MSK-IMPACT used in the corpus.

  • PMID:36862133 — MSK-IMPACT (up to 505 genes) with select targeted RNA sequencing was used in the Make-an-IMPACT direct-to-patient program; 250/333 enrolled patients had tumor DNA of sufficient quality. In histiocytosis (n=84), most commonly mutated actionable genes were BRAF (33%), MAP2K1 (13%), KRAS (7%), and CSF1R (2.4%) PMID:36862133.
  • PMID:37315267 — MSK-IMPACT 410–505 was used in 40% of 61 extremity rhabdomyosarcoma patients; identified mutually exclusive CDK4 amplifications and CDKN2A deletions associated with poor OS PMID:37315267.
  • PMID:37591896 — one of the panel versions used in the 233-patient NSCLC brain metastasis cohort PMID:37591896.
  • PMID:39289779 — IMPACT505 was used to profile 733 CSF samples (later cohort, Nov 2018 – Nov 2022) in a 1,007-sample CSF ctDNA study at MSK; ctDNA positivity was associated with a three-fold increased risk of death (HR 3.23, p < 0.001) and 50.7% of ctDNA-positive samples carried a level 1 OncoKB actionable alteration PMID:39289779.
  • PMID:39753968 — IMPACT505 was one of four panel generations (n=345 patients) used in the 2,336-patient MSK PDAC cohort; KRAS mutant-allele dosage gains detected with FACETS (v0.5.14) correlated with worse OS across all clinical stages independent of whole-genome doubling PMID:39753968.
  • PMID:37699004 — IMPACT505 (up to 505 genes) used to sequence 902/1,123 esophagogastric cancer patients at MSK; germline analysis used a 76–88-gene MSK-IMPACT panel on 466 patients PMID:37699004.
  • PMID:37769223 — IMPACT505 used in 37 (72.5%) of 51 HCC patients for matched tumor tissue profiling, complementing MSK-ACCESS cfDNA profiling; 92.5% of tissue alterations were detected in matched cfDNA PMID:37769223.
  • PMID:38630790 — IMPACT505 used in an additional 105-patient validation cohort for the GNH subset of diffuse pleural mesothelioma; 4 GNH cases identified PMID:38630790.
