GZMA

Overview

GZMA encodes granzyme A, a serine protease released by cytotoxic lymphocytes.

Alterations observed in the corpus

  • Effector cytotoxic component of the 20-gene ICR signature used in the AC-ICAM colon cancer atlas PMID:37202560.
  • Component of the cytolytic-activity signature (geometric mean of GZMA + PRF1); elevated expression in tumor RNA of metastatic melanoma patients with clinical benefit from ipilimumab CTLA-4 blockade (P = 0.042) PMID:26359337
  • No significant differential expression between anti-PD-1 responders vs non-responders in whole-tumor transcriptome analysis of pretreatment melanoma biopsies (bulk RNA-seq may dilute immune-cell-specific signal) PMID:26997480
  • GZMA geometric mean with PRF1 defines the cytolytic activity (CYT) score; on-therapy CYT increases associated with benefit in both ipilimumab-naive (Ipi-N, p=0.005) and ipilimumab-progressed (Ipi-P, p=0.043) melanoma patients treated with nivolumab PMID:29033130
