STAT1
Overview
STAT1 is a Th1 signaling transcription factor included in the 20-gene Immunologic Constant of Rejection (ICR) immune signature.
Alterations observed in the corpus
- STAT1 is a component of the 20-gene ICR panel (Th1 signaling genes) used to stratify colon cancer immune subtypes PMID:37202560.
- STAT1 is an IFN-stimulated gene (ISG) upregulated from the earliest HGSOC precursor stage (p53.I incidental fimbriae) onward in fallopian tube epithelium, alongside IFITM1, IRF7, IRF9, ISG15, TAP1, and MX1; upregulation is activated by the cGAS–STING–TBK1 axis triggered by micronuclear rupture in pre-malignant cells. PMID:39386723
Cancer types (linked)
- COAD — ICR panel component; contributes to ICR-high vs ICR-low prognostic stratification (HR 0.54, P=0.0095) PMID:37202560.
- High-grade serous ovarian carcinoma (HGSOC) precursor lesions — STAT1 is a constituent of the interferon-response gene program activated in p53 signatures, STICs, and invasive HGSOC; upregulation begins in the earliest pre-malignant fallopian tube lesions (p53.I). PMID:39386723
Co-occurrence and mutual exclusivity
- Co-expressed with other ICR Th1 genes (IFNG, TBX21, CD8A, IRF1) as a module PMID:37202560.
Therapeutic relevance
- No direct targeted therapy reported in the corpus; ICR module may have predictive utility for immunotherapy PMID:37202560.
Open questions
- None specific to STAT1 in the corpus.
